Introduction of Maize Controlling Element " Ac " into Petunia hybrida

Abstract

|
We have transformed Petunia hybrids (cv. White Madness) with Agrobacterium tumefaciens that have non-tumorigenic T DNA carrying the maize transposable element Activator $quot;Ac$quot;. After 2-3 weeks of inoculation with Agrobacterium, the calli were formed in callus inducing media supplemented with 100 ㎎/ℓ kanamycin. The petunia leaves mostly produced multi-shoots. Transformation was assured by checking the stability on media supplemented with kanamycin. Integration and copy number of the Ac element in the transgenic plant were confirmed by Southern blot analysis.
